Recovery and ampli®cation of plasmid DNA with atomic force microscopy and the polymerase chain reaction

چکیده
Mapping the force on plasmid DNA molecules with the atomic force microscope (AFM) showed an increased adhesion force between a silicon nitride tip and DNA strands in a mildly acidic solution. With this adhesion force, the plasmid DNA could be stretched off the mica surface, and the silicon nitride tip occasionally picked up one or several molecules of plasmid DNA from the mica surface. We have been able to develop a polymerase chain reaction (PCR) method, with which one molecule of plasmid DNA could be speci®cally and ef®ciently ampli®ed, to prove the presence of plasmid DNA on the silicon nitride AFM tip. It opens up a possibility for recovering one or several DNA molecules with an AFM tip from biological sample surfaces including that of a chromosome. # 1998 Elsevier Science B.V.

EVALUATION OF DIRECT MICROSCOPY, CULTURE, AND POLYMERASE CHAIN REACTION FOR THE DIAGNOSIS OF TUBERCULOUS MENINGITIS
In view of the importance of early diagnosis of tuberculous meningitis (TBM), the efficiency of the polymerase chain reaction (PCR), one of the most reliable and sensitive DNA-based assays, was compared with conventional methods (acid-fast microscopy and culture) for the detection of M. tuberculosis in cerebrospinal fluid (CSF) specimens from patients suspected of TBM. Of the 29 CSF specim...
متن کاملDNA studies using atomic force microscopy: capabilities for measurement of short DNA fragments
Short DNA fragments, resulting from ionizing radiation induced DNA double strand breaks (DSBs), or released from cells as a result of physiological processes and circulating in the blood stream, may play important roles in cellular function and potentially in disease diagnosis and early intervention. The size distribution of DNA fragments contribute to knowledge of underlining biological proces...
متن کاملPolymerase chain reaction method for the rapid detection of virulent Shigella spp.
Bacillary dysentery, or shigellosis, is a disease of humans in which the colonic epithelium is invaded by bacteria and subjected to inflammatory destruction. The aim of this study was to develop a polymerase chain reaction(PCR) test for detection of virulent Shigella spp.. For this purpose, the primers were designed to amplify a 526-bp internal region of the Shigella spp. icsA gene, which encod...
متن کاملDetection of single Dactylogyrus spp. in DNA extracted from infected gill tissue of fishes using Polymerase Chain Reaction
Dactylogyrus spp. are monogenean worms found mostly as ectoparasites on the gills of several fish species, including carp and goldfish. These parasites are commonly detected by microscopic analysis of the gill lamellae, but this is time-consuming and technically difficult. In contrast to this conventional method, molecular techniques provide specific, sensitive and safe detection of parasites. ...

The real-time quantitative polymerase chain reaction (rtqPCR) has overcome the limitations of conventional, time-consuming quantitative PCR strategies and is maturing into a routine tool to quantify gene expression levels, following reverse transcription (RT) of mRNA into complementary DNA (cDNA). Expression pro®ling with single-cell resolution is highly desirable, in particular for complex tis...
